Endothelial dysfunction underlies the pathogenesis of multiple sclerosis, in
which there is massive infiltration of autoreactive immune cells through damaged cerebral endothelium into the central nervous system. Leukocyte-endothelial interaction is a dynamic process dependent on many endogenous factors, which role and mechanisms of involvement are not fully understood. Using an in vitro model, we studied the effect of ionotropic glutamate receptor
ligands on the viability of HUVECs, as well as on their expression of cell adhesion molecules PECAM-1 and VCAM-1. Glutamate was found to increase endotheliocytes viability, with glycine, an NMDA-receptor coagonist, enhancing the
effect of glutamate. Blockade of AMPA-receptors is accompanied by a decrease
in this index. The analysis showed the direct participation of NMDA-receptors in
the regulation of surface expression of VCAM-1 adhesion molecule. The results
indicate the involvement of the glutamatergic system as an important regulator
of leukocyte-endothelial interaction.
